A Study on Histological Changes in the Gonads, and Changes in Content of water and Electrolytes in the Tissue of the Uterus and Stomach of Immture Rabbits by Chlorpromazine Administration


Abstract
Chlorpromazine is known that to be a marked antiadrenergic and local anesthetic properties, although it possesses mild anti-cholinergic and anti-histaminic actions, as well as a sedative, anti-emetic and hypothermic central actions.
In view of theories on the site of action of chlorpromazine, endocrinological changes were reported such as inhibition of gonadal stimulation, amenorrhea and breast swelling accompanied by lactation. The author has studied the effect of chlorpromazine to the gonads and changes in content of water and electrolytes in the uterus and stomach of immature female rabbits, around 1.0kg.
A group of rabbits treated with 50 I. U. /kg. of HCG and another group with 50mg. /kg. of chlorpromazine plus 50 I. U. /kg. of HCG were observed 24 hours after conditioning.
Measurment of Na and K was carried out by Flamephotometry, Schales and Schales method for Cl content and Kovacs and Tarrnoky¢¥s method for Ca and Mg contents.
The results were as follows;
1. The HCG treated rabbits showed remarkable acceleration in maturation of the follicles and ovula tion in the ovaries while the chlorpromazine plus HCG treated group showed no obvious suppression in ovulation.
2. k and Mg in the serum of the HCG treated rabbits showed increase in level but decreased contents of Cl, Na and Ca. On the contrary, chlorpromazine plus HCG treated group, K and Mg were decreased abut Ca was increased in its level.
3. Water and Cl in the uterine tissue of the HCG treated rabbits showed increase in content compared to the normal group but Na, K and Mg were decreased in each level. In the chlorpromazine plus HCG treated group, water, Cl and Mg were decreased, K and Na were increased when compared to only HCG treated group.
4. Water and Cl in the stomach tissue of the HCG treated rabbits showed increase in contents and Na, Ca and Mg were decreased but there was no changes in K level. In the chlorpromazine plus HCG treated group, water, Cl and Mg were decreased, but there was no changes in Na, K and Ca levels.
5. The content of water in the stomach tissue of the normal, HCG, chlorpromazine plus HCG treated group were increased than that of the uterine tissue.
6 The Cl in the stomach tissue of the normal and HCG treated group showed decrease in level compared with the uterine tissue but increased in chlorpromazine plus HCG treated group.
7. The contents of K, Ca and Mg in the stomach tissue of the normal, HCG and chlorpromazine plus HCG treated group were increased than that of the uterine tissue but Na in the stomach tissue showed lower levels than in the uterine tissue.
